Non-crushable pill formulations and methods of using the formulations are disclosed. A non-crushable pill formulation for preventing unintended use of a drug, comprising a polymer, the polymer forming a polymer backbone of the complex; cross-linkers, the cross-linkers connecting the polymer backbone through covalently bonding to form at least one inner cavity within the complex; and the drug, the drug being trapped either covalently or non-covalently in the at least one inner cavity within the complex, wherein the drug is protected from releasing outside of the complex.

COMPOSITIONS FOR CONTROL OF DRUG ABUSE
Opiates, amphetamines, barbiturates and other drugs such as benzodiazepines are extensively abused or misused and are frequently the cause of death by overdosing. These drugs are also prone to oxidation and the final degradation products depend on the reactants and the reaction conditions. This invention describes the use of inactivating agents such as permanganates, peroxides, persulfates, bismuthates, periodates or other oxidants in a dosage form as an approach to minimize abuse and overdose. The product is designed such that the inactivating agent is released if there is an attempt to extract the drug from the formulation or in cases of overdose. Once released, the inactivating agent quickly degrades the drug and converts it into inactive compounds. Since the reactants (drug and inactivating agent) are incompatible in situations of normal drug usage, they are kept separated within the vehicle of the invention, but released for interaction in case of misuse. A catalyst may be included in the formulation to facilitate the reaction.
Regimen for supressing organ rejection
The present invention relates to a method of suppressing organ rejection in a patient receiving an organ transplant by initiating oral treatment with a once-daily extended release tacrolimus dosage form, for example, at an initial dose of from about 0.15 to about 0.20 mg/kg/day within 24 or 48 hours following transplantation. The once-daily extended release tacrolimus dosage form (i) provides low fluctuation and/or swing of tacrolimus, (ii) provides a significantly lower C.sub.max than an immediate release formulation of tacrolimus while providing the same or greater area under the curve (AUC), (iii) releases the tacrolimus substantially in the colon and/or the lower ileum, (iv) releases at most 63.5% of the tacrolimus in the dosage form at the 12 hour time point, or (v) any combination of any of the foregoing.

The disclosure is directed to pharmaceutical compositions for oral administration in form of coated tablets that exhibit modified release properties when administered as either whole or half tablets. In particular, the disclosure is directed to modified release tablets comprising deferiprone, said tablets being suitable for twice daily oral administration. The disclosure is also directed to methods of making and using the same.
Solid preparation composition for oral administration of colonic purgative containing anhydrous sodium sulfate, potassium sulfate, anhydrous magnesium sulfate and simethicone
The present invention relates to a solid colonic purgative for oral application, containing anhydrous magnesium sulfate, potassium sulfate, anhydrous sodium sulfate, and, additionally, simethicone. The present invention is more satisfactory in the colon cleansing effect even with the doses reduced by up to about 20 percent compared to those of a conventional colonic purgative consisting of anhydrous magnesium sulfate, potassium sulfate and anhydrous sodium sulfate. Besides, the present invention, due to the reduced doses, has less unpleasant taste or odor caused by the main ingredients and requires a less intake of the preparation and water, improving drug compliance significantly. Further, unlike the conventional solid colonic purgative, the present invention can be prepared into tablets without using a water-soluble lubricant. In other words, the present invention can be formulated into a solid preparation for oral application by using a water-soluble lubricant, or even without using any water-soluble lubricant.
