A temperature control device (2) comprises a number of active thermal sites (6) disposed at respective locations on a substrate (10), each comprising a heating element (13) for applying a variable amount of heat to a corresponding site of a medium and a thermal insulation layer (16) disposed between the heating element and the substrate. At least one passive thermal region (8) is disposed between the active thermal sites (6) on the substrate (10), each passive thermal region (8) comprising a thermal conduction layer (18) for conducting heat from a corresponding portion of the medium to the substrate (10). The thermal conduction layer (18) has a lower thermal resistance in a direction perpendicular to a plane of the substrate (10) than the thermal insulation layer (16). This enables precise control over both heating and cooling of individual sites in a flowing fluid, for example.

A temperature control method for the thermal cycling device can control whether liquid contacts the temperature adjustment device through the liquid delivery device in coordination with whether the temperature adjustment device heats the thermally-conductive carrier, so as to produce rapid heating and heat dissipation effects.

Intermittent warming of a biologic sample including a nucleic acid includes receiving at a first end of a channel of a microfluidic device, a biologic sample including a nucleic acid, and warming a subset of a plurality of heating elements disposed adjacent to the channel. The method includes warming the heating elements to a particular temperature of a particular warming and cooling protocol. The method includes moving the biologic sample from the first end of the channel to a second end of the channel opposite the first end at a particular flow rate associated with the warming and cooling protocol, and intermittently warming the biologic sample using the subset of heating elements while the biologic sample moves from the first end of the channel to the second end of the channel.

A sonicator assembly, including: a microplate defining a plurality of wells; a manifold for containing a transducer fluid that is thermally coupled to the plurality of wells of the microplate; an ultrasonic generator operable for applying an ultrasonic excitation to the wells of the microplate; one or more of a heating module thermally coupled to and operable for selectively heating the transducer fluid and a cooling module thermally coupled to and operable for selectively cooling the transducer fluid; and a controller operable for controlling operation of the ultrasonic generator and the one or more of the heating module and the cooling module. The controller is further operable for monitoring a temperature and a pressure within the manifold. A temperature of the plurality of wells is controllable over a temperature range from 4° C. to 95° C. Optionally, the plurality of wells include a plurality of heat-resistant round-bottom hydrophilic wells.

TEMPERATURE CONTROLLED REACTOR

A reactor is provided for controlling the temperature of materials undergoing a physical or chemical process. The reactor includes a reactor having a holder adapted to hold a plurality of the materials in a plurality of 2×n arrays, each array having two rows of n materials. The reactor further includes a fluid inlet adapted to receive a temperature-modifying fluid and a fluid outlet adapted to discharge the temperature-modifying fluid. The sample holder has a plurality of fluid flow channels configured such that a fluid flow channel is located adjacent to each of the two rows in each of the plurality of 2×n arrays. The fluid flow channels are adapted to provide substantially equal flow rates and substantially equal fluid flow volumes as the temperature-modifying fluid travels past the arrays. The sample holder may also be provided with a plurality of inlet-side air chambers, each inlet-side air chamber disposed between the fluid inlet and a respective one of the arrays and adapted to thermally insulate the materials from the temperature-modifying fluid as it is introduced into the fluid inlet.
