POLYPEPTIDE FRAGMENT B (MP-B) AND USE THEREOF

A polypeptide fragment B (MP-B) has an amino acid sequence shown in SEQ ID NO: 1, in which an amino acid Xaa at position 10 is Arg, Phe, Glu, Thr, or absent, an amino acid Xaa at position 16 is Asn, Val, Leu, Gly, or absent, and an amino acid Xaa at position 25 is Ser, Glu, Met, Arg, or absent. The MP-B can significantly improve the colonic pathologic morphology and decrease a colonic histopathologic score and an expression level of colonic interferon-γ (IFN-γ) in an inflammatory bowel disease (IBD) mouse model, and shows the ability to interfere with the occurrence of IBD in mice.

Probiotic Bacterial Molecules and their Use in Methods to Treat/Prevent Infection by Harmful Bacteria and to Provide Nutritional Health

This invention provides isolated and characterized secreted molecules from probiotic bacteria for use in compositions and methods for the treatment and/or prevention of infection by harmful pathogenic bacteria. The isolated secreted molecules can also be used in nutritional or medical food products which provide probiotics to the gastrointestinal tract of a mammal.

The present invention relates to the treatment of diseases relating to proteins of the human microbiome metasecretome and, thus, to microbiome interactions, especially microbiome-host interactions. In particular the present invention relates to a method for identification of secreted peptides and proteins of the human microbiome. The present invention also relates to methods for generating a database of human microbiome metasecretome protein sequences. Furthermore, the present invention relates to a method for preparing a protein of the human microbiome metasecretome as well as to the use of such proteins in medicine.

POLYPEPTIDE FRAGMENT D (MP-D) AND USE THEREOF

A polypeptide fragment D (MP-D) has an amino acid sequence shown in SEQ ID NO: 1, in which an amino acid Xaa at position 9 is Tyr, Val, Gly, Ser, Gln, or absent, an amino acid Xaa at position 20 is Ser, Gln, Glu, Tyr, Arg, or absent, and an amino acid Xaa at position 25 is Asn, Thr, Ser, Pro, Leu, or absent. The MP-D can significantly improve the colonic pathologic morphology and decrease a colonic histopathologic score and an expression level of colonic interferon-γ (IFN-γ) in an inflammatory bowel disease (IBD) mouse model, and shows the ability to interfere with the occurrence of IBD in mice.

A polypeptide fragment C (MP-C) has an amino acid sequence shown in SEQ ID NO: 1, in which an amino acid Xaa at position 9 is Tyr, Val, Gly, Ser, or Gln, an amino acid Xaa at position 20 is Ser, Gln, Glu, or Tyr, an amino acid Xaa at position 30 is Asn, Thr, Ser, Pro, or Leu, and an amino acid Xaa at position 42 is Gly, Arg, Met, or absent. The MP-C can significantly improve the colonic pathologic morphology and decrease a disease activity index (DAI) and a colonic histopathologic score in an inflammatory bowel disease (IBD) mouse model, and shows the ability to interfere with the occurrence of IBD in mice.

Artificial secretion peptides for heterologous protein production

Provided herein, in some embodiments, are artificial secretion peptides capable of directing secretion from Lactobacillus for use, for example, in producing heterologous proteins, including therapeutic proteins.

Process for the linear synthesis of gram-positive class II bacteriocins and compositions and uses thereof

A process for the linear synthesis of a gram-positive class II bacteriocin or a variant thereof is disclosed herein. The process comprises the stepwise addition of selected amino acids to a solid support; pseudoproline positioning and reopening; and cleavage of the gram-positive class II bacteriocin or the variant thereof from the solid support to provide a linear gram-positive class II bacteriocin or variant thereof; and in situ disulfide bond formation. Various applications and uses of the synthetic bacteriocins are also disclosed. The synthetic process can also be used to synthesize variants of bacteriocins by the selective substitution of one or more amino acids and/or additions and/or deletions of selected amino acids.
