Hydrogels from dynamic covalent networks

In an embodiment is provided a polymer that includes a plurality of N-J-N or N—C—S repeating units, wherein each J is independently a carbon atom, an alkyl group, or an aryl group; a plurality of hydrophilic groups bonded with the repeating units; and a plurality of hydrophobic groups bonded with the hydrophilic groups and the repeating units. In another embodiment is provided hydrogels of such polymers. The hydrogels may be used as delivery vehicles for various payloads. In another embodiment is provided methods of forming such polymers.

The disclosed technology provides a thermoplastic copolymer comprising: a plurality of difunctional triketone species; (b) a plurality of a first diamine species, wherein the first diamine species contains one or more primary amine groups and/or one or more secondary amine groups, and wherein the first diamine species does not contain a tertiary amine group; a plurality of a second diamine species, wherein the second diamine species contains one or more primary amine groups and/or one or more secondary amine groups, wherein the second diamine species does not contain a tertiary amine group, and wherein the second diamine species is different than the first diamine species; and optionally, a plurality of monofunctional amine-reactive groups. Some embodiments provide segmented thermoplastic copolymers. Methods of making and using the thermoplastic copolymer are also described, including depolymerizing the thermoplastic copolymer to form recycled monomers. The recycled monomers may then be repolymerized in a closed-loop system.

Polymer for delivery of biologically active materials

The present invention mainly relates to a polymer for delivery of biologically active materials, a complex and a method of synthesis thereof. The polymer comprises a poly(ethylene imine) and at least one monomer, each monomer comprising a modified sugar moiety, preferably galactose, comprising a sulphur atom or a nitrogen atom and a chemical moiety comprising a terminal epoxide for linking the polyethylene imine to the monomer, wherein the sulphur atom or the nitrogen atom links the modified sugar moiety to the chemical moiety. The biologically active material is preferably a gene, siRNA, mRNA or plasmid DNA. Further disclosed is the medical use of said complex in treating a disease caused by a genetic disorder, for example cancer.

Class of sequence-defined polymers and preparation methods thereof

The invention provides a new class of synthetic sequence-defined polymer (SDP) and a method of synthesizing the same. The synthetic sequence-defined polymers have dithiocarbamate incorporated to the backbone. The method introduces a functional group dithiocarbamate in the backbone by using a new support-free, protection-deprotection free three-component reaction strategy. Dithiocarbamate-SDP is prepared from a unique bifunctional monomer, CS.sub.2 and chloroacetyl chloride. Chloracetyl chloride is used as a co-monomer. Different functional groups may be introduced in the dithiocarbamate-SDP via custom synthesis of monomers with the desired functional group, using the method disclosed. The SDPs may undergo modular post-synthetic modification through multiple paths. SDP is produced in multi-gram scale at low cost and in an eco-friendly manner through the method. No hazardous waste is produced in the process as HCl gas released from the reaction may be neutralized by bicarbonate in the medium.
