The present invention discloses an in-vitro culture, induction, activation and cryopreservation method and cell bank establishment for immune cells. The method includes the follows: using a dedicated amplification medium of immune cells to perform first-stage amplification culture on mononuclear cells to obtain preliminarily amplified immune cells; using a dedicated induction medium of immune cells to perform second-stage induction and amplification culture on the preliminarily amplified immune cells to obtain induced immune cells; using a dedicated activation medium of immune cells to perform third-stage activation and amplification culture on the induced immune cells to obtain a large number of immune cells with activation functions; using a dedicated cryopreserving fluid of immune cells to cryopreserve the immune cells to obtain cryopreserved immune cells; and performing preservation according to ABO/RH typing and HLA typing; and establishing an information file of immune cells for retrieval to construct an immune cell bank.

Disclosed are means, methods and compositions of matter for treatment Parkinson's Disease through concurrent immune modulation and regenerative means. In one embodiment Parkinson's Disease is treated by augmentation of T regulatory cell numbers and/or activity while concurrently providing regenerative cells such as mesenchymal stem cells, and/or dopamine secreting cells. In one embodiment administration of immunoglobulins such as IVIG together with low dose interleukin-2 and/or low dose naltrexone is disclosed as a preparatory means prior to administration of therapeutic cells such as stem cells. Other therapeutic means utilized in an adjuvant manner are also provided for hormonal rebalancing, transcranial magnetic stimulation, and deep brain stimulation.

The improved 4-5 day, optionally 3-5 day GMP-compliant in-vitro method enables the production of macrophages from monocytes that benefits from a shorter cell culture time, fewer interventions whilst maintaining the desired characteristics of the human macrophages. The present invention describes a method wherein the monocytes are cultured in medium comprising one or more growth actors to stimulate macrophages with a pro-regenerative phenotype. The method described herein is xeno-free, serum-free and GMP compliant. In addition, further disclosed are macrophages produced according to the present invention and the use of said macrophages in the treatment of liver diseases, such as liver cirrhosis.
SEED CELL MEDIUM OF TUMOR-INFILTRATING LYMPHOCYTE AND APPLICATION THEREOF
A seed cell medium of a tumor-infiltrating lymphocyte and an application thereof. The medium contains a cell culture component, a cell factor, and an immune checkpoint antibody or an antigen-binding fragment thereof. The cell factor includes IL-2; an immune checkpoint includes PD-1, LAG-3, TIGIT, and/or CTLA-4; and the cell culture component is a serum medium or a serum-free medium. The seed cell medium accelerates a culture of a seed cell of the tumor-infiltrating lymphocyte and shortens the amplification time required by the tumor-infiltrating lymphocyte.

The present invention is targeted towards reinvigorating exhausted Tumor Infiltrating Lymphocytes (TILs) in vitro by co-culturing excised TIL containing tumor fragments with checkpoint inhibitors, stimulating the TILs with other interleukins known to revert T cell exhaustion), and/or inhibiting the effect of regulatory T cells secreted factors (such as IL-10) thereby creating a favorable tumor microenvironment (TME) where exhausted T-cells can expand faster and to higher numbers than currently established TIL expansion protocols.
