METHODS FOR PRODUCTION OF PLATELETS FROM PLURIPOTENT STEM CELLS AND COMPOSITIONS THEREOF

Methods for production of platelets from pluripotent stem cells, such as human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs) are provided. These methods may be performed without forming embryoid bodies or clusters of pluripotent stem cells, and may be performed without the use of stromal inducer cells. Additionally, the yield and/or purity can be greater than has been reported for prior methods of producing platelets from pluripotent stem cells. Also provided are compositions and pharmaceutical preparations comprising platelets, preferably produced from pluripotent stem cells.

CELL REPROGRAMMING METHOD

The present invention pertains to a method for creating reprogramed cells from somatic cells without gene introduction. The method includes a step (a) for culturing somatic cells in a medium containing a histone deacetylase inhibitor, and a step (b) for culturing the cells cultured in step (a) in a medium containing an OCT3/4 transcription stimulating factor to create reprogrammed cells.

The method comprises the following steps: isolating cells from the urine, and then culturing the cells with a culture medium of urine-derived kidney stem cells on feeder cells to obtain the urine-derived kidney stem cells, wherein the feeder cells are fibroblasts, and the culture medium of urine-derived kidney stem cells contains 200-300 mL of DMEM medium, 200-300 mL of F12 medium, 20-70 mL of fetal bovine serum, 0.2-2 mM of L-glutamine, 1-14 ng/mL of insulin, 0.1-1 ng/mL of epidermal growth factor, 5-30 μg/mL of adenine, and 2-20 μg/mL of hydrocortisone. By using the method, kidney stem cells with high proliferation capacity and specificity can be obtained and applied, and thus the regenerative outcome of the kidney tissue after injury can be improved.

Provided are new strategies, methods and systems, described herein as vasoactive intestinal peptide (VIP)-assisted air-liquid-interface (ALI) culture, to significantly increase the number of enteroendocrine (EEC) and enterochromaffin (EC) cells over the traditional submerged culture, while at the same time maintaining a high barrier integrity of monolayers. The new strategies, methods and systems overcome the limitations of the existing EEC enrichment methods by maintaining high cell viability and barrier integrity and without requiring complicated procedures of cocultures or genetic engineering/induction. The created EEC-enriched, contiguous monolayer platform acts as a robust analytical tool to enable functional studies of hormone secretion from EEC cells with high signal background ratio and repeatability.
