The present application relates to composition of matter, processes and use of composition of matter relating to flavivirus proteins, peptides, and epitopes, for example, for therapeutic or preventative vaccination against one or more flavivirus serotypes or species, and/or for inducing, enhancing, or sustaining an immune response against at least one flavivirus serotype or species. The flavivirus may be for example the Zika and/or Dengue virus.

A biosensor molecule comprises: a protease amino acid sequence; at least one sensor comprising at least one sensor amino acid sequence which is responsive to at least one target molecule; and an inhibitor of the protease activity of said protease amino acid sequence; wherein the biosensor is switchable from a protease active to a protease inactive state, or from a protease inactive to a protease active state when said sensor responds to said target molecule. The biosensor protease may be a protease of a virus such as a Potyvirus or a Flavivirus wherein the inhibitor is an autoinhibitory peptide derived from the virus. The biosensor may respond to the target molecule allosterically or may be cleaved by a target protease molecule.

ZIKA VIRUS VACCINES

Disclosed is an immunogen comprising a fusion protein, wherein the fusion protein comprises a Zika virus (ZIKV) envelope protein, optionally a signal peptide, and a multimerization domain. The signal peptide is a premembrane (prM) signal peptide, an IgG signal peptide, or a human secretory signal peptide hidden Markov model, and the multimerization domain is an immunoglobulin Fc domain, a T4 fibritin foldon trimerization domain, or a human collagen XV trimerization domain. Nucleic acids, vectors, and microneedle arrays including these compositions are disclosed. Methods of producing an immune response to ZIKV are also disclosed.
