The invention relates to a unit dose of a dengue vaccine composition and methods and uses for preventing dengue disease and methods for stimulating an immune response to all four dengue virus serotypes in a subject or subject population. The unit dose of a dengue vaccine composition includes constructs of each dengue serotype, such as TDV-1, TDV-2, TDV-3 and TDV-4, at various concentrations in order to improve protection from dengue infection.

The present invention is directed to a method of booster vaccination and to a vaccine composition for use in such a method, for inducing in a human subject a neutralizing antibody response, wherein said subject has previously received a primary vaccination against each of serotypes 1 to 4 of dengue virus and was dengue naïve before said primary vaccination, said composition comprising a dengue antigen of at least one of serotypes 1 to 4 or a nucleic acid construct capable of expressing said antigens in the subject, wherein said booster vaccination results in a 2-fold increase in the neutralizing antibody titre against each of serotypes 1 to 4. The invention is also directed to a method of inducing in a human subject a neutralizing antibody response comprising the administration of a vaccine composition, or to a vaccine composition for use in such a method, said composition comprising a dengue antigen of each of serotypes 1 to 4, or a nucleic acid construct capable of expressing in said subject a dengue antigen of each of serotypes 1 to 4; wherein said composition is administered as a primary vaccination, followed by a booster vaccination, and wherein the human subject is initially dengue naïve.

Specific resurfaced Dengue virus glycoprotein subunit E DIII variants and their uses in preventing and treating Dengue virus infection are disclosed. Provided herein are specific resurfaced Zika virus glycoprotein subunit E DIII variants and their uses in preventing and treating Zika virus infection. Multivalent vaccines comprising such resurfaced EDIII variants are also described, in addition to nanoparticle conjugated resurfaced EDIII variants.
Inactivating pathogens and producing highly immunogenic inactivated vaccines using a dual oxidation process
Provided are surprisingly effective methods for inactivating pathogens, and for producing highly immunogenic vaccine compositions containing an inactivated pathogen rendered noninfectious by exposure to a Fenton reagent, or by exposure to a Fenton reagent or a component thereof in combination with a methisazone reagent selected from the group consisting of methisazone, methisazone analogs, functional group(s)/substructure(s) of methisazone, and combinations thereof. The methods efficiently inactivate pathogens, while substantially retaining pathogen antigenicity and/or immunogenicity, and are suitable for inactivating pathogens, or for the preparation of vaccines for a wide variety of pathogens with genomes comprising RNA or DNA, including viruses and bacteria. Also provided are highly immunogenic inactivated vaccine compositions prepared by using any of the disclosed methods, and methods for eliciting an immune response in a subject by administering such vaccine compositions.
ZIKA VIRUS IMMUNOGENIC COMPOSITIONS
Provided herein are immunogenic compositions for the prevention of Zika virus infections. Disclosed herein are immunogenic compositions comprising an expression vector and a nucleotide sequence disposed therein, wherein the nucleotide sequence comprises: a nucleotide sequence encoding a Zika virus NS3 protein. Further disclosed herein are methods for preventing a Zika virus infection in a subject in need thereof, the method comprising administering a therapeutically effective amount of an immunogenic composition of the present disclosure to the subject.
DENGUE VIRUS VACCINE COMPOSITIONS AND METHODS OF USE THEREOF
The present invention relates to dengue virus vaccine compositions comprising a first and a second dengue vaccine, wherein the first dengue vaccine comprises at least one live, 5 attenuated dengue virus or live, attenuated chimeric dengue virus and the second dengue vaccine is a recombinant dengue subunit vaccine, a DNA vaccine, a conjugate vaccine, or an inactivated dengue vaccine; wherein the genome of the live attenuated dengue virus or the live attenuated chimeric dengue virus comprises a 30 nucleotide deletion of the TL2 stem-loop structure of the 3′ untranslated region. The dengue virus vaccine compositions of the invention may further 10 comprise one or more adjuvants. In preferred embodiments of the invention, the first and the second dengue vaccine are tetravalent. The invention also relates to methods of using the dengue virus vaccine compositions of the invention to treat or prevent dengue infection, or to prevent, ameliorate, or delay the onset or progression of the clinical manifestations thereof.
