Attenuated live vaccine against mycoplasmal pneumonia of swine (MPS) and use thereof

Disclosed are an attenuated live vaccine against mycoplasmal pneumonia of swine (MPS) and use thereof. In the present invention, pathological lung tissues of swine having typical Mycoplasma hyopneumoniae (Mhp) infection and no obvious other pathogenic infections are screened, and subcultured 100 generations in lungs of newborn rabbits; then, Mhp strains are isolated and serially subcultured in a medium; and the Mhp strain AN306 is obtained by screening a plurality of strains, which is deposited with an accession number: CCTCC NO. M2012431. Also disclosed is a live vaccine formulation against MPS prepared on the basis of the attenuated strain and comprising live attenuated strain, a pharmaceutically acceptable carrier or excipient, and optionally an adjuvant and immunogens of other pathogens.

MYCOPLASMA PNEUMONIAE IMMUNOASSAY METHOD AND IMMUNOASSAY INSTRUMENT

An immunoassay utilizing a monoclonal antibody which specifically reacts with both of P1 protein and P30 protein of Mycoplasma pneumoniae, which monoclonal antibody has a higher affinity with P1 protein and P30 protein of Mycoplasma pneumoniae than the known monoclonal antibodies, is disclosed. The immunoassay utilizes an antigen-antibody reaction between a monoclonal antibody which specifically reacts with P1 protein and P30 protein of Mycoplasma pneumoniae and which specifically reacts with a peptide containing an amino acid sequence composed of PPQPG or antigen-binding fragment thereof, and P1 protein and P30 protein derived from Mycoplasma pneumoniae.

A method for testing presence or absence of a Mycoplasma in a specimen is disclosed. The method includes: culturing a specimen; extracting a nucleic acid from the cultured specimen and performing amplification of the nucleic acid using a primer capable of amplifying a nucleic acid sequence derived from a Mycoplasma, in which the specimen is a biotechnology-related product or a part thereof.

It also relates to the use of M. hyopneumoniae as a host for expressing recombinant proteins and other DNA sequences of interest.

Also provided are compositions and methods for administration to birds of the order Galliformes.
