Methods of Library Preparation
Disclosed herein is a modified transposon end sequence comprising a mosaic end sequence, wherein the mosaic end sequence comprises one or more mutation as compared to a wild-type mosaic end sequence, wherein the mutation comprises a substitution with a uracil, an inosine, a ribose, an 8-oxoguanine, a thymine glycol, a modified purine, or a modified pyrimidine. Also disclosed are transposome complexes comprising these modified transposon end sequences and methods of library preparation using these modified transposon end sequences.

This disclosure relates to methods and compositions for detecting a target nucleic acid sequence using a dual-hapten probe. More specifically, the present disclosure relates to using recombinase polymerase amplification (RPA) and a dual-hapten probe to detect a target nucleic acid sequence. In some cases, the detection is on lateral flow strips.
Method for Reducing Sequencing Errors Caused by DNA Fragmentation
Provided herein, among other things, is a method for reducing sequencing errors caused by mechanical DNA fragmentation. In some embodiments, this method may comprise: (a) obtaining a DNA sample; (b) mechanically fragmenting the DNA sample to produce a template; (c) treating the fragmented DNA with a plurality of DNA repair enzymes; and (d) obtaining a sequence of the fragmented DNA. The treatment step (c) reduces the number of fragmentation induced errors.
