DETAILED DESCRIPTION OF EMBODIMENTS

[0035] When a complex formulation formed by comprising colchicine and metformin as effective agents provided in embodiments of the present invention is administered to obese rats, an effect of improving obesity is confirmed in the obese rats and the present invention has been completed based thereon.

[0036] Hereinafter, various embodiments of the present invention will be described in detail.

[0037] The colchicine as the main effective agent used herein is a material extracted from a plant of colchicum genus and a drug used mainly as an alternative drug for the gout when a nonsteroidal anti-inflammatory agent does not smoothly exhibit the therapeutic effect on the gout, and is used even for the treatment of familial Mediterranean fever, periceritis, Behcet's disease, etc.

[0038] The metformin as another effective agent used herein has a blood glucose improvement effect as a biguanide-based drug and is mainly used as a therapeutic agent for diabetes. The mechanism of metformin is not accurate, but is known to prevent an increase in concentration of cAMP induced by glucagons by inhibiting cellular respiration occurring in mitochondria and activating AMPK and consequently inhibit the activity of PKA to inhibit the production of glucose.

[0039] The inventors of the present invention have confirmed through various experiments that therapeutic efficacy for obesity is increased when co-administering colchicine and metformin.

[0040] In accordance with one exemplary embodiment of the present invention, when comparing changes in body weight of rats in the case of single administration of colchicine, single administration of metformin, and co-administration of colchicine and metformin to obesity-induced rats, a weight loss effect of rats is greatest in the case of co-administration of colchicine and metformin.

[0041] Further, in accordance with one exemplary embodiment of the present invention, when comparing expression levels of obesity-related genes SREBP-1c and FAS (fatty acid synthase) in the case of single administration of colchicine, single administration of metformin, and co-administration of colchicine and metformin to obesity-induced rats, the expression levels of the genes are reduced greatest in the case of co-administration of colchicine and metformin.

[0042] Sterol regulatory element binding proteins (SREBPs) genes are transcription factors that regulate homeopronability and metabolism of lipids, and these genes precisely regulate the expression of enzymes required for synthesis of endogenous cholesterol, fatty acids (FA), triasyl glycerol (TG) and phospholipids.

[0043] The SREBPs include three types of 1a, 1c and 2, and it has been known that SREBP-1a and SREBP-1c are mainly involved in the synthesis of fatty acids and triglycerides and SREBP-2 is involved in the cholesterol metabolism. In the liver tissue, the expression of SREBP-1c is predominant and regulates the expression of genes such as FAS, ACC, stearoyl-CaP desaturase, ATP citrate lyase, and malic enzyme associated with the synthesis of triglycerides in hepatocytes.

[0044] Hyperinsulinemia due to insulin resistance increases the biosynthesis of fatty acids by increasing the expression of SREBP-1c in the liver and consequently causes the accumulation of triglycerides in liver tissue. The role of SREBP-1c in the insulin resistance and the accumulation of triglycerides in the liver has been proven through animal experiments. Through studies of reporting that when fatty liver lesions are observed in ob/ob mice, as a feature of high obesity and insulin resistance and the SREBP-1 gene is inactivated in obesity-induced rats, the accumulation of triglycerides in the liver tissue is reduced by about 50%, it can be seen that SREBP-1c plays an important role in the occurrence of fatty liver in an insulin-resistant animal model.

[0045] The SREBPs are involved in many diseases causing processes such as ER stress, inflammation, autophagocytosis and apoptosis, and are known to cause obesity, dyslipidemia, diabetes, non-alcoholic fatty liver disease, etc.

[0046] When administering the complex formulation provided in embodiments of the present invention, the colchicine is added in an amount of specifically 5 to 300 μg/kg, more specifically 50 to 200 μg/kg, and the metformin is added in an amount of specifically 50 to 400 mg/kg, more specifically 100 to 200 mg/kg.

[0047] When the content of the colchicine is less than 5 μg/kg, the efficacy of the colchicine is not almost exhibited, and when the content thereof is more than 300 μg/kg, there is a side effect that causes gastrointestinal disorders at the time of taking.

[0048] When the content of the metformin is less than 50 mg/kg, the efficacy thereof is not exhibited, and when the content thereof is more than 400 mg/kg, side effects such as abdominal pain, diarrhea, vomiting, etc. are likely to occur.

[0049] Further, in the pharmaceutical composition for treating adipositas provided in embodiments of the present invention, the content ratio of the colchicine and metformin as the effective agents may include 1:1,000 to 1:20,000, more specifically 1:2,000 to 1:5,000 as the content of metformin to the content of colchicine.

EXAMPLE

[0058] Preparation of Experimental Animals and Experimental Method

[0059] Low fat diet (LFD: containing 10 kcal % fat) or high fat diet (HFD: containing 60 kcal % fat) feed was taken autonomously for 12 weeks in 6-week-old C57BL/6J sp. male rats. The obesity-induced rats due to the intake of the HFD feed additionally took the HFD feed for 12 weeks and were administered with colchicine of 50, 100, and 200 μg/kg or metformin of 200 mg/kg alone or co-administered with the colchicine and the metformin. A control drug, Xenical (orlistat ingredient) was administered alone at 60 mg/kg. All drugs were orally administered once a day.

[0060] Changes in body weight of obesity-induced rats were measured according to single administration of colchicine or metformin and co-administration of colchicine and metformin, which were illustrated in FIG. 1.

[0061] As illustrated in FIG. 1, it was confirmed that an increase in body weight induced by the HFD was not changed by single administration of colchicine and the body weight was slightly reduced in single administration of metformin. In comparison, in co-administration of colchicine and metformin, it was confirmed that the weight loss of obesity-induced rats was much higher than single administration of the two ingredients, and particularly, when the content of colchicine was smaller than the content of metformin, a weight loss effect was relatively excellent.

[0062] This effect was shown to be more significant than the control drug, Xenical.

[0063] After the drugs were administered to the experimental animals, obesity-induced rats, the intakes of the feed were compared, and as illustrated in FIG. 2, there was no large difference in the intakes of the feed for each drug.

[0064] Next, after the drugs were administered to the obesity-induced rats, the weights of epididymal fat were measured and the results thereof was illustrated in FIG. 3.

[0065] As illustrated in FIG. 3, it was confirmed that the weight of epididymal fat of obesity-induced rats increased by the HFD was not changed by single administration of colchicine and slightly reduced in single administration of metformin. In comparison, in co-administration of colchicine and metformin, it was confirmed that the weight of epididymal fat was much reduced, and particularly, when the content of colchicine was smaller than the content of metformin, it was confirmed that the weight loss effect was relatively excellent, and much more reduced than the control drug, Xenical.

[0066] Meanwhile, after the drugs were administered to the obesity-induced rats, the expression of obesity-related genes SREBP-1c and FAS was measured, and the results thereof were illustrated in FIGS. 4A and 4B.

[0067] According to FIG. 4, in the epididymal fat of the obesity-induced rats by the HFD, it was confirmed that the expression of the obesity-related genes SREBP-1c and FAS was not changed by single administration of colchicine and slightly reduced by single administration of metformin. In comparison, in co-administration of colchicine and metformin, it was confirmed that the expression of SREBP-1c and FAS was much reduced in the epididymal fat of the obesity-induced rats. Particularly, it was confirmed that when the content of colchicine was smaller than the content of metformin, the expression of the genes was relatively much reduced, and much more reduced than the control drug, Xenical.

[0068] Finally, after the drugs were administered to the obesity-induced rats, changes in blood levels of aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured and illustrated in Table 1 below.

[0069] As illustrated in Table 1, it was confirmed that the blood levels of AST and ALT increased by the HFD were not changed by single administration of colchicine and slightly reduced in single administration of metformin. In comparison, in co-administration of colchicine and metformin, it was confirmed that the blood levels of AST and ALT were much reduced, and it was confirmed that when the content of colchicine was smaller than the content of metformin, the levels were much reduced and much more reduced than the control drug, Xenical.

TABLE-US-00001 TABLE 1 Group ALT (U/L) AST (U/L) ND 32.55 ± 1.03  35.41 ± 1.51 HFD 305.16 ± 2.05  408.58 ± 1.36 HFD + Colchicine (50 μg/kg) 175.08 ± 2.78  372.26 ± 3.75 HFD + Colchicine (100 μg/kg) 164.5 ± 2.11 346.91 ± 3.90 HFD + Colchicine (200 μg/kg) 157.38 ± 3.3   309.24 ± 2.61 HFD + Metformin (200 mg/kg) 156.41 ± 2.77  284.37 ± 2.78 HFD + Metformin (200 mg/kg) + 54.98 ± 1.84  61.44 ± 2.65 Colchicine (50 μg/kg) HFD + Metformin (200 mg/kg) + 92.45 ± 1.25 127.63 ± 3.27 Colchicine (100 μg/kg) HFD + Metformin (200 mg/kg) + 125.50 ± 3.43  153.41 ± 3.42 Colchicine (200 μg/kg) HFD + Orlistat (60 mg/kg) 175.20 ± 3.11  203.31 ± 3.72

[0070] As seen from the results, there was almost no effect of reducing the fat of the obesity-induced rats by single administration of colchicine, and there was a slight effect by single administration of metformin, but the effect was significantly shown by co-administration of colchicine and metformin. This means that the two drugs exhibit an apparent synergic effect to efficiently prevent and treat the obesity, and the pharmaceutical composition of combining colchicine and metformin provided in the present invention may be used as an excellent pharmaceutical composition for treating or preventing the obesity.
