Universal vaccine based on shared tumor neoantigens for prevention and treatment of micro satellite instable (MSI) cancers
This invention relates to a method of selecting a collection of frame-shift peptides (CFSPs) to produce a universal cancer vaccine peptide collection (CVP) for prophylaxis and treatment of patients with hereditary and sporadic micro-satellite instability (MSI) tumors. This invention relates as well to a method of producing a CVP by selecting a subset of frame-shift peptides (FSPs) from the CFSP and optionally modifying the FSP's amino acid (aa) sequence to generate modified FSPs (mFSPs). The invention further relates to nucleic acid collections encoding a CVP of FSPs and/or mFSPs in one or more vaccine vectors that can be used also simultaneously. These CVPs, nucleic acids and vectors are used for the prophylaxis or treatment of MSI cancers.
Chromatography ligand comprising domain C from <i>Staphylococcus aureus </i>protein A for antibody isolation
The present invention relates to a chromatography ligand, which comprises Domain C from Staphylococcus protein A (SpA), or a functional fragment or variant thereof. The chromatography ligand presents an advantageous capability of withstanding harsh cleaning in place (CIF) conditions, and is capable of binding Fab fragments of antibodies. The ligand may be provided with a terminal coupling group, such as arginine or cysteine, to facilitate its coupling to an insoluble carrier such as beads or a membrane. The invention also relates process of using the ligand in isolation of antibodies, and to a purification protocol which may include washing steps and/or regeneration with alkali.

The present invention relates to a compound of formula (I) AA-AA-AA-R.sub.1—R.sub.2 (I) wherein, in any order, 2 of said AA (amino acid) moieties are cationic amino acids and 1 of said AA is an amino acid with a lipophilic R group, the R group having 14-27 non-hydrogen atoms; R.sub.1 is a N atom, which may be substituted by a branched or unbranched C.sub.1-C.sub.10 alkyl or aryl group, which group may incorporate up to 2 heteroatoms selected from N, O and S; and R.sub.2 is an aliphatic moiety having 2-20 non-hydrogen atoms, said moiety being linear, branched or cyclic. The invention further relates to formulations containing these compounds, solid supports having these compounds attached thereto, the use of these compounds in therapy, particularly as antimicrobial, anti-tumour or anti-biofilm agents and non-therapeutic uses of these compounds, particularly their use in inhibiting biofilm formation or removing a biofilm.

The present application addresses the problem of providing an Fc-binding protein having an improved antibody separation ability. The present application also addresses the problem of providing a high-accuracy antibody separation method using an insoluble carrier having the protein immobilized thereon. The problems can be solved by: an Fc-binding protein in which at least an amino acid substitution at a specific position therein occurs and which has reduced affinity for an antibody; and an antibody separation method including allowing an equilibration buffer solution to pass through a column in which an insoluble carrier having the protein immobilized thereon is filled to equilibrate the column, adding a solution containing an antibody to cause the adsorption of the antibody onto the carrier, and eluting the antibody adsorbed on the carrier using an elution solution.

The fusion protein or conjugated protein has a partial peptide comprising at least seven consecutive amino acid residues of an amino acid sequence encoded by a predetermined DNA, and a ligand directly or indirectly bound to the partial peptide and having the capability of binding to cell surfaces. The ligand is preferably an antibody. The intracellular delivery carrier comprises the fusion protein or conjugated protein. The cell membrane permeation enhancer comprises the partial peptide.
