Identification of protective antigenic determinants of porcine reproductive and respiratory syndrome virus and uses thereof
The invention relates to a polypeptide of a protective antigenic determinant (PAD polypeptide) of porcine reproductive and respiratory syndrome virus (PRRSV) and nucleic acids encoding a PAD polypeptide. The PAD polypeptide and nucleic acids encoding a PAD polypeptide are useful in the development of antibodies directed to PAD, vaccines effective in providing protection against PRRSV infection, and diagnostic assays detecting the presence of PAD antibodies generated by a PAD-specific vaccine. The invention also discloses methods of generating antibodies to PAD, for vaccinating a pig to provide protection from PRRSV infections, a method of preparing the vaccine, a method of treating PRRSV infections in a pig, and a method of detecting antibodies to PAD of PRRSV.
Needle-free administration of PRRSV vaccines
The invention provides novel methods and compositions for the vaccination of porcine animals against porcine reproductive and respiratory syndrome virus (PRRSV). Described herein are immunological and/or vaccine compositions comprising a DNA vector encoding a PRRSV protein, particularly a truncated ORF7 protein, which are administered to porcines using needle-free delivery. The plasmid can include more than one nucleic acid molecule such that the plasmid can express more than one antigen. Also disclosed are methods for using and kits employing such compositions.
Antigen-surface-coupled liposome vaccine for non-human animals
Disclosed is a vaccine for non-human animals, which vaccine enables effective immune induction in non-human animals such as livestock or poultry. The vaccine for non-human animals of the present invention comprises liposomes each comprising an antigen molecule(s) bound to the surface thereof, the antigen molecule(s) being derived from a pathogen infectious to the non-human animals. The non-human animals are, for example, livestock or poultry. One most preferred example of the vaccine according to the present invention is an infectious bronchitis virus (IBV) vaccine, which is effective against various isolated IBV strains, and which is expected to be sufficiently applicable even to cases where a mutant strain appeared. Another preferred example of the vaccine according to the present invention is a PRRSV vaccine, which was confirmed to have an effect that reduces fever, lung lesion development, and the like caused by the infection. The present invention enables development of liposome vaccines effective for prevention of infections caused by various non-human animal pathogens including PRRSV as well as IBV.
