Method for determining optimal preservation temperature of anaerobic ammonium oxidation biofilm in wastewater treatment for total nitrogen removal
The present disclosure discloses a method for determining optimal preservation temperature of the anaerobic ammonia oxidation biofilm in wastewater treatment, and belongs to the technical field of environmental engineering. The method of the present disclosure characterizes the ratio of living cells, early apoptotic cells, late apoptotic cells and dead cells in the anaerobic ammonia oxidation biofilm by flow cytometry, and the optimum storage temperature can be measured within a few hours. The method of the present disclosure performs correlation analysis on the characteristic indexes of the anaerobic ammonia oxidation biofilm activity recovery process to verify the reliability of the data. By using the method of the present disclosure, the step of recovering the biofilm activity can be omitted, the removal rates of ammonia nitrogen and total nitrogen were over 90% and 85%, respectively.

Disclosed herein include systems, devices, and methods for determining a gating strategy. An acquisition system can cause a particle analyzer to collect parameter measurements of some particles of a sample. An analysis system can receive the parameter measurements and determine a gating strategy from the parameter measurements. The acquisition can collect measurements of some or all of the remaining particles of the sample using the gating strategy determined by the analysis system.
Kill event optimization
A method of choosing which undesired cell to destroy in a multi-cell fluorescent event includes detecting fluorescence of cells, converting photons detected in the fluorescence into an analog voltage output signal, and identifying at least two discernable peaks associated with the cells. By looking solely at properties measured within the multi-cell fluorescent event, a decision of which cell to target for elimination can be made. Using this method with large population sizes can result in an effective sex skewed product. The sex skewed product can, for example, be formed from bull semen which is then later used to inseminate cows which results in an increased likelihood of giving birth to female cattle.
Parallel single cell lens free optical dielectrophoresis cytometer
A dual light source lens-free dielectrophoresis (DEP) flow cytometer for massively parallel single cell analysis. Each cells dielectric is inferred from measuring their altitude and subsequently velocity change due to DEP actuation in a microfluidic channel. Dual LED sources facilitate velocity measurement by producing two shadows for each cell passing through the channel. These shadows are detected using a linear optical array detector. Massively parallel analysis is possible as each pixel of the detector can independently analyze the passing cells. The DEP cytometer is composed of simple modular components and has the potential to be scaled to achieve a significantly high throughput label-free single-cell analyzer.

Aspects of the present disclosure include methods for modulating an intensity profile of a laser beam. Methods according to certain embodiments include irradiating an acousto-optic device with a laser to generate an output laser beam having a plurality of angularly deflected laser beams, capturing an image of the output laser beam, determining an intensity profile of the output laser beam along a horizontal axis from the captured image and adjusting one or more parameters of a waveform inputted into the acousto-optic device in response to the determined intensity profile to generate an output laser beam having a modulated intensity profile. Systems having a laser, an acousto-optic device, an imaging sensor and a waveform generator as well as non-transitory computer readable storage medium with instructions for practicing the subject methods are also described.
HIGH-CONTENT ANALYSIS METHOD
The present invention relates to a method for subjecting a plurality of microwells containing cells to a high-content assay, said method comprising: a) Acquiring at least one image of said plurality of microwells; b) In said image, detecting a plurality of areas of interest, each area of interest corresponding to a single cell; c) Measuring at least one derived property, and, optionally, at least one direct property of said areas of interest, where said one or more properties is a selection property; d) Selecting a subset of said plurality of microwells, where said microwells belonging to the subset contain areas of interest selected based on said at least one selection property; e) Extrapolating an output parameter from a property measured in the set of areas of interest selected, where said property is defined as output property, said output property being distinct from said selection properties where said output parameter is the processing of an output property measured in said set of areas of interest. In a further aspect, there are claimed a system for subjecting a plurality of microwells containing cells to a high-content assay and a computer program which comprises instructions for subjecting a plurality of microwells containing cells to a high-content assay.
CHIP FIXING DEVICE AND PARTICLE INSPECTION DEVICE
A chip fixing device includes a mounting portion on which a channel chip is mounted and a fixing unit for fixing the channel chip mounted on the mounting portion. The channel chip includes a channel through which a liquid containing a particle flows and a pressure changing unit for introducing the particle of interest from the channel. The mounting portion includes a substrate on which the channel chip is set. The fixing unit includes a fixing member for pressing the channel chip against the substrate, a piezoelectric element for actuating the pressure changing unit, and a holding member configured to hold the piezoelectric element and movable in a direction in which the holding member approaches and separates from the pressure changing unit. The fixing member includes an elastically deformable portion for fixing the holding member to the fixing member by deforming elastically when pressing the channel chip against the substrate.
Estimating Oocyte Quality
The present invention extends to methods, systems, and computer program products for estimating oocyte quality. A machine learning algorithm accesses oocyte training data for a mammalian species (e.g., humans) and trains a neural network to estimate oocyte quality for the mammalian species based on the oocyte training data. The neural network accesses a microscopic image of an oocyte and identifies oocyte features of the oocyte. Based on the identified oocyte features, the neural network estimates oocyte quality, including: (a) predicting a probability of a corresponding embryo maintaining sufficient developmental competence until a specified time after fertilization and (b) predicting another probability of the corresponding embryo reaching a specific embryonic stage after fertilization. An oocyte is selected, from among a plurality of human oocytes including the human oocyte, for a potential recipient based at least in part on the oocyte quality, including based on the probability and the other probability.
