Embodiments of this invention include methods for detecting in vitro the presence of Memory B cells in peripheral blood mononuclear cells (PBMCs) that produce antibodies reactive to CNS antigens associated with Multiple Sclerosis (MS). Stimulating PBMCs from patients with MS by a polyclonal stimulating agent promotes the ability of B-lymphocytes, when exposed to a CNS antigen, to produce antibodies specific for the CNS antigen. In contrast, stimulating PBMCs from subjects without MS do not produce such responses.
Antibody-dependent cell-mediated phagocytosis assay for reliably measuring uptake of aggregated proteins
The present disclosure provides methods of assaying for antibody-dependent cell-mediated phagocytosis (ADCP). In some embodiments, the methods include monomerizing and labeling a protein, contacting the protein with a protein-specific antibody to form an antibody-protein complex, contacting the antibody-protein complex with a phagocytic cell to permit phagocytosis, and assessing the amount of internalized fluorescence.
Sorting biological and non-biological moieties using magnetic levitation
Systems and methods for levitating populations of moieties, cells, or other such units using one or more magnets in a microfluidic environment are provided. These systems and methods may be used to, for example, separate or sort heterogeneous populations of the units from one another, to assembly a multi-unit assembly during the levitating of the units, and to evaluate samples at the point of care in real-time. These systems and methods may also utilize a frame that enables an imaging device, such as a smartphone, to capture the units in real time as they are manipulated in the system.
Methods for detection of rare subpopulations of cells and highly purified compositions of cells
Methods are provided for detection of a target cell type within a cell population, and compositions are provided comprising cells and an indicator that indicates the number of cells of the target cell type in the cell population. Examples are provided in which these methods are used to detect human embryonic stem cells within a differentiated cell population with exquisite sensitivity. Differentiated cells produced from embryonic stem cells can be characterized by these methods before transplantation into a recipient, thereby providing further assurance of safety.
Identification and isolation of human corneal endothelial cells (HCECs)
The present invention provides methods for the identification, isolation and/or enrichment of human corneal endothelial cells (HCECs). In some embodiments, the method comprises a positive selection process in which a cell population containing human corneal cells is contacted with a positive affinity reagent that selectively binds to HCECs relative to cells other than HCECs (e.g., corneal keratocytes, etc.) in the population and/or a negative selection process in which a cell population containing HCECs is contacted with a negative affinity reagent that selectively binds to cells other than HCECs in the population relative to HCECs. The present invention also provides reagents and kits for the identification, isolation and/or enrichment of HCECs as well as compositions that are enriched in HCECs.

Adult stem cell compositions and methods of identification and isolation
Methods, compositions and cells are provided that identify and isolate a population of adult non-embryonic progenitor cells having multilineage potential, physical diameters of about 2 μm to about 8 μm in size or about 4 μm to about 6 μm, and expressing at least one of the stem cell associated genes among Oct-4, KLF-4, Nanog, Sox-2, Rex-1, GDF-3 or Stella. Methods are also provided that identify and isolate populations, which are subsets or subpopulations of progenitor adult stem cells within the population of the adult stem cells which is a heterogeneous population, the methods including contacting the adult stem cells with a ligand specific for at least one of: CD99, tetraspan, ICAM4, full-length MUC1, and truncated MUC1 receptor, in which a presence of a surface protein on the cells that bind to the ligand identifies the population which is the subset of the differentiated progenitor adult stem cells.
