Method for determining the in vivo interaction mode
Herein is reported a method for determining the binding interaction with a multimeric antigen of an antibody of the human IgG1 subclass that has at least two binding sites specifically binding to the antigen comprising the steps of 1) determining the binding affinity of the antibody for the multimeric antigen, and 2) incubating a mixture comprising the antibody and a polypeptide that is derived from lysine-gingipain of Porphyromonas gingivalis under conditions and for a time sufficient to cleave the antibody into Fabs and Fc-region, and determining the binding affinity of the Fabs of the antibody for the multimeric, whereby the binding affinity of the antibody to the multimeric antigen to be affinity-driven if the binding affinity determined in both steps are comparable and to be avidity-driven if the binding affinity determined in both steps are different.

The present invention is an intraoral examination method that, for example, assesses the state of periodontal disease and/or caries by measuring the levels of bacteria present in an intraoral sample from an intraoral sample collected from a subject.
SALIVA PAIRED IMAGING TEST APPARATUS AND METHOD
An apparatus and method for remote saliva testing in a variety of environments. The test involves the healthcare provider and the patient. Included is a non-invasive method of saliva collection and result detection which allows healthcare providers to send test kits to patient homes for remote testing, monitoring and care decisions. The apparatus includes a dental saliva imaging test card paired with a web application downloaded to a smartphone. The system may be used at home or in the dental office. The smartphone interface walks patients and providers through the test with tutorials to accurately capture testing results. The imaging test card includes a plurality of test strips onto which the patient places saliva. The test strips are sensitized to look for certain biomarkers, some of which detect periodontal disease and elevated caries risk. Results from the test strips are photographed and are then uploaded to the application for analysis.
DIRECT AFFINITY MEASUREMENT OF HUMAN IGG1 BINDING MULTIMERIC ANTIGENS
Herein is reported a method for determining the binding affinity of the binding sites of a bivalent full length antibody of the human IgG1 subclass to a homo-multimeric antigen comprising the steps of i) incubating a mixture comprising the antibody and a polypeptide that is derived from lysine-gingipain of porphyromonas gingivalis at a pH of from pH 7.5 to pH 8.5, in the presence of a reducing agent, at a temperature of from 30 C. to 42 C., for time of from 10 min. to 240 min. to cleave the antibody into Fabs and Fc-region, and ii) determining the binding affinity of the Fabs of the antibody for its antigen using a surface plasmon resonance method by directly applying the incubated reaction mixture obtained in the previous step in the surface plasmon resonance method and therewith determining the binding affinity of the binding sites of the bivalent full length antibody of the human IgG1 subclass.

The in vitro diagnostic method for periodontal disease of the present invention includes bringing a sample derived from an oral cavity of a subject into contact with an electrode to perform electrochemical measurement in the presence of a glucogenic amino acid and an electron mediator under an anaerobic environment and providing information for judging that periodontal disease is progressing in the oral cavity of the subject when current generation is detected as a result of the electrochemical measurement. According to the in vitro diagnostic method for periodontal disease of the present invention, it is possible to provide information for judging the progress of periodontal disease by simple operation.
