Antibody constructs and methods of treating cancer

Provided herein are antibodies and related polypeptides that bind specifically to CEACAM6. The antibodies and/or polypeptides can be configured as bispecific T cell engagers. The antibodies and/or polypeptides can also be configured as chimeric antigen receptors. Also provided are methods of detection and treatment of cancer, for example, pancreatic cancer, using the antibodies and related polypeptides herein.

Disclosed herein are methods of diagnosing or prognosing aggressive prostate cancer in a subject and methods of treating a subject with aggressive prostate cancer. For example, the methods can include measuring increased expression of aggressive prostate cancer-related molecules (such as FOLH1, SPARC, TGFB1, CAMKK2, NCOA2, EGFR, or PSA) and optionally administering a therapeutically effective amount of aggressive prostate cancer therapy.

Provided are methods, assays, and kits for detecting hepatocellular carcinoma, as well as methods for stratifying subjects among higher and lower risk categories for having hepatocellular carcinoma, and methods of treating and managing treatment of subjects that are suspected or at risk of having hepatocellular carcinoma. Although previous work has attempted to address the need for a highly sensitive, early predictor of hepatocellular carcinoma by assessment of one or more biological factors, none have approached the degree of sensitivity that is required for clinically relevant determination of whether a subject, especially a non-symptomatic subject, has that condition. The present inventors have discovered that certain combinations of factors fulfill this need by conferring a high level of accuracy that was not previously attainable.

MECHANISTIC BIOMARKER FOR PREDICTING THE SURVIVAL OF PANCREATIC CANCER PATIENTS

The present disclosure provides biomarkers and methods of use thereof for diagnosing and prognosing pancreatic cancer and other cancers in a subject. The biomarkers comprise protein kinase C (PKC), PH domain and leucine rich repeat protein phosphatase 1 (PHLPP1), and the ratio of PKC/PHLPP1 and can be detected using anti-PKC and/or anti-PHLPP1 antibodies and quantified using immunochemistry techniques. Methods of treating pancreatic cancer with PHLPP1 inhibitors are also provided.
