NEWCASTLE DISEASE VIRUS-BASED VECTORED VACCINE
20220402975 · 2022-12-22 ·

Provided are compositions and methods for vaccinating against picornaviruses. The compositions include modified Newcastle Disease viruses (NDVs) that are sufficient to produce virus-like particles (VLPs) in a host recipient. The modified NDVs contain a single stranded negative sense RNA polynucleotide having nucleotide sequences configured in a 3′-5′ direction encoding sequentially NDV nucleocapsid protein (NP), phosphoprotein (P), matrix protein (M), fusion protein (F), hemagglutinin-neuraminidase (HN) and RNA-dependent RNA polymerase (L) protein. A first nucleotide sequence encoding a picornavirus capsid polyprotein precursor is positioned between the between P and M nucleotide sequences. A second nucleotide sequence encoding a picornavirus protease that is capable of processing the capsid polyprotein precursor is positioned between the HN and L nucleotide sequences. Purified, infectious non-pathogenic NDV particles are included, as are methods for using such particles for vaccination against any infectious picornavirus. Kits and articles of manufacture containing and/or for making the NDV particles are also provided.

An immunogen includes an antigenic EGFRvIII peptide linked to a Qβ bacteriophage virus-like particle (VLP) carrier. The antigenic EGFRvIII peptide has at least 53%, at least 61%, at least 69%, at least 76%, at least 84%, or at least 92% sequence similarity amino acid similarity to LEEKKGNYVVTDH (SEQ ID NO:1).

Virus and antigen purification and conjugation

Disclosed herein are methods of forming compounds and exemplary compounds in the nature of a conjugated compound demonstrating enhanced stability, which in some embodiments comprises a protein and virus particle mixed in a conjugation reaction to form a conjugate mixture, such that the conditions and steps of forming these products allow for unrefrigerated storage for longer time periods than previous approaches, thus making feasible access to such products over a global supply chain.

METHODS AND COMPOSITIONS FOR INHIBITING POLYOMAVIRUS-ASSOCIATED PATHOLOGY

Disclosed herein are methods of eliciting an immune response against a polyomavirus (for example, BKV serotype I (BKV-I), BKV serotype II (BKV-II), BKV serotype III (BKV-III) and/or BKV serotype IV (BKV-IV)) and methods of treating or inhibiting polyomavirus-associated pathology (such as polyomavirus-associated nephropathy, BKV-associated hemorrhagic cystitis, or JC virus-associated progressive multifocal leukoencephalopathy; PML). Further disclosed are immunogenic compositions of use in the disclosed methods. Also disclosed are methods of selecting an organ transplant donor and/or recipient including detecting whether the prospective donor and/or recipient has BKV serotype-specific (such as BKV serotype IV-specific) neutralizing antibodies.

ONCOLYTIC VIROTHERAPY AND IMMUNOTHERAPY

The present disclosure concerns combination therapy for cancer thatutilizes (i) an oncolytic virus; (ii) a virus comprising nucleic acid encoding an immunomodulatory factor; and (iii) at least one cell comprising a chimeric antigen receptor (CAR) specific for a cancer cell antigen. In particular embodiments, the virus comprises nucleic acid encoding an immunomodulatory factor comprises nucleic acid encoding IL-12 and/or antagonist anti-PD-L1 antibody.
