DEVICES AND METHODS FOR DETERMINING NUCLEIC ACIDS USING DIGITAL DROPLET PCR AND RELATED TECHNIQUES
The present disclosure generally relates, in certain aspects, to droplet-based microfluidic devices and methods. In certain aspects, target nucleic acids contained within droplets are amplified within droplets in a first step, where multiple primers may be present. However, multiple primers may cause multiple target nucleic acids to be amplified within the droplets, which can make it difficult to identify which nucleic acids were amplified. In a second step, the amplified nucleic acids may be determined. For example, the droplets may be broken and the amplified nucleic acids can be pooled together and sequenced. As an example, new droplets may be formed containing the amplified nucleic acids, and those nucleic acids within the droplets amplified by exposure to certain primers.
HIGH-LEVEL MULTIPLEXING REACTION VESSEL, REAGENT SPOTTING DEVICE AND ASSOCIATED METHODS
Reaction vessels, cartridges, devices and methods for facilitating high-level multiplexing are described herein. Such reaction vessels can include a planar frame defining a fluidic path between a first planar substrate and a second planar substrate, a fluidic interface is located at one end of the planar frame with a pair of fluidic ports, a well chamber and a pre-amplification chamber. Devices for spotting reagents in wells of high-level multiplexing reaction vessels and improved reagent solutions are also described herein.

Disclosed is a measurement apparatus including: a support mechanism configured to support a cartridge in which a chamber is formed, the chamber being configured to store a measurement sample that generates light an intensity of which varies depending on an amount of a test substance; a photodetector configured to detect the light generated from the measurement sample stored in the chamber; and a reflection member provided between the photodetector and the cartridge supported by the support mechanism, the reflection member having an inner face, the reflection member being configured to reflect, at the inner face, the light generated from the measurement sample stored in the chamber, and guide the light to the photodetector, wherein the reflection member is configured to have an area surrounded by the inner face, the area decreasing from a side where the cartridge supported by the support mechanism is provided toward a side where the photodetector is provided.
CHEMILUMINESCENCE MEASUREMENT APPARATUS
Disclosed is a chemiluminescence measurement apparatus that includes: a support member configured to support a cartridge for measuring a test substance contained in a specimen by chemiluminescence measurement; a motor configured to rotate the support member so as to rotate the cartridge such that a process required for the chemiluminescence measurement proceeds in the cartridge; and a light receiver configured to receive light generated by chemiluminescence in the cartridge that is supported by the support member rotated by the motor. The cartridge supported by the support member and a light receiving surface of the light receiver are disposed inside a dark space surrounded by a light-shielding portion, and the motor is disposed outside the dark space.

Dissolution guided wetting of structured surfaces
A microfabricated device having at least one gas-entrapping feature formed therein in a configuration that entraps air bubbles upon wetting the feature with a solvent or solution is described. The device includes a sacrificial residue in contact with the gas-entrapping feature, the dissolution of which guides the wetting of the gas-entrapping feature.
Systems and methods for biomimetic fluid processing
Systems and methods generating physiologic models that can produce functional biological substances are provided. In some aspects, a system includes a substrate and a first and second channel formed therein. The channels extend longitudinally and are substantially parallel to each other. A series of apertures extend between the first channel and second channel to create a fluid communication path passing through columns separating the channels that extends further along the longitudinal dimension than other dimensions. The system also includes a first source configured to selectively introduce into the first channel a first biological composition at a first channel flow rate and a second source configured to selectively introduce into the second channel a second biological composition at a second channel flow rate, wherein the first channel flow rate and the second channel flow rate create a differential configured to generate physiological shear rates within a predetermined range in the channels.
