Well-plate incubator

Incubators are disclosed which include an enclosure with an internal chamber configured to support a cell culture plate and provide an environment suitable for maintaining and/or culturing biological cells. The enclosure can include one or more openings configured to allow access to the cell culture plate. The incubators can further include a structure having a plurality of openings configured to be aligned with a corresponding plurality of wells in the cell culture plate, and a sealing element configured to moveably seal the plurality of openings in the structure. The sealing element can comprise a plurality of openings corresponding to at least a subset of the plurality of openings of the structure. Access to the internal chamber can be provided by aligning the plurality of openings in the sealing element with the plurality of openings in the structure. Methods for using the incubators are also provided.

It is proposed that the first coupling device (5) has a fluid inlet (7) and a fluid outlet (8) and also a first cannula (9), the latter having an end which is directed away from the fluid inlet (7) and forms the fluid outlet (8), that the second coupling device (6) has a fluid passage (10) which, in an initial state of the sterile connector (1), is covered axially by a first septum (11), that the fluid outlet (8), in the initial state of the sterile connector (1), is arranged in a hermetically sealed region of the first coupling device (5) and the surface (11a) of the first septum (11) facing axially away from the fluid passage (10) is arranged in a hermetically sealed region of the second coupling device (6), and that, in a fluidic connection process starting from the initial state of the sterile connector (1), a fluidic connection between the fluid inlet (7) and the fluid passage (10) can be produced by the fact that the end of the first cannula (9) forming the fluid outlet (8) pierces the first septum (11) of the second coupling device (6).

A method and apparatus of aseptic processing and production of cells in a non-sterile enclosure apparatus without chemical biocides is disclosed, by controlling the level of humidity throughout the enclosure to 25% relative humidity (RH) or less, and preferably 20% or 15% or less RH. In addition, the temperature is controlled to 37° C., and consistent gas flow is maintained the enclosure. Colony forming units from microbial contamination detected by environmental monitoring within the enclosure are significantly reduced in this method.
