Systems and methods to produce B cells genetically modified to express selected antibodies

Systems and methods to genetically modify B cells to express selected antibodies are described. The systems and methods can be used to: obviate the need for classical vaccinations; provide protection against infectious agents for which no vaccinations are currently available; provide protection against infectious agents when patients are otherwise immune-suppressed; and/or provide a benefit provided by a therapeutic antibody, such as in the treatment of autoimmune disorders.

GENE TRANSFER SYSTEM
20230009161 · 2023-01-12 ·

The present disclosure relates to a viral gene delivery vector particle and a bispecific polypeptide configured to bind a viral gene delivery vector particle and target cell-specific receptor protein. The disclosure also relates to gene delivery systems, compositions, and methods of use thereof.

NEUTRALIZING GP41 ANTIBODIES AND THEIR USE

Monoclonal neutralizing antibodies are disclosed that specifically bind to the HIV-1 gp41 membrane-proximal external region (MPER). Also disclosed are compositions including the disclosed antibodies that specifically bind gp41, nucleic acids encoding these antibodies, expression vectors including the nucleic acids, and isolated host cells that express the nucleic acids. The antibodies and compositions disclosed herein can be used for detecting the presence of HIV-1 in a biological sample, or detecting an HIV-1 infection or diagnosing AIDS in a subject. In additional, the broad neutralization breadth of the disclosed antibodies makes them ideal for treating a subject with an HIV infection. Thus, disclosed are methods of treating and/or preventing HIV infection.

HIV VACCINE COMPOSITIONS, METHODS, AND USES THEREOF

The present invention discloses immunogenic compositions including recombinant peptides and proteins comprising human immunodeficiency viruses (HIV) antigens and immunogens, e.g., gp 120 protein peptides. In some aspects, the immunogenic composition comprises a secreted fusion protein comprising a soluble HIV viral antigen joined by in-frame fusion to a C-terminal portion of a collagen which is capable of self-trimerization to form a disulfide bond-linked trimeric fusion protein. In some aspects, the immunogenic compositions provided herein are useful for generating an immune response, e.g., for treating or preventing an HIV infection. In some aspects, the immunogenic compositions provided herein may be used in a vaccine composition, e.g., as part of a prophylactic and/or therapeutic vaccine. Also provided herein are methods for producing the recombinant peptides and proteins, prophylactic, therapeutic, and/or diagnostic methods, and related kits.

BROADLY NEUTRALIZING ANTIBODIES AGAINST HIV

The present invention provides broadly neutralizing antibodies against HIV, compositions comprising the same and methods of use thereof.

Combinations of Vaccines and Neutralizing Antibodies for Treating Human Immunodeficiency Virus Infection in Subjects Undergoing Antiretroviral Treatment

Methods for inducing an immune response against Human Immunodeficiency Virus (HIV) in HIV-infected subjects undergoing antiretroviral therapy (ART) are described. The methods involve initial administration of an adenovirus vector vaccine and subsequent administration of a poxvirus vector vaccine, followed by administration of anti-HIV broadly neutralizing antibodies (bNAb).

Artificial secretion peptides for heterologous protein production

Provided herein, in some embodiments, are artificial secretion peptides capable of directing secretion from Lactobacillus for use, for example, in producing heterologous proteins, including therapeutic proteins.

OPTIMIZED CROSSLINKERS FOR TRAPPING A TARGET ON A SUBSTRATE

The presently-disclosed subject matter relates to crosslinkers, compositions, and methods for trapping a target of interest on a substrate of interest. The methods may be used to inhibit and treat pathogen infection and provide contraception. The methods may be used to trap or separate particles and other substances. The subject matter further relates to methods of identifying and preparing optimal crosslinkers and methods for manipulating targets of interest.

NK ENGAGER COMPOUNDS THAT BIND VIRAL ANTIGENS AND METHODS OF USE

This disclosure describes compounds that engage NK cells and methods of using the compounds. Generally, the compound includes an NK engaging domain, a targeting domain that selectively binds to a target cell, and an NK activating domain operably linking the NK engaging domain and the targeting domain. In an illustrative embodiment, the targeting domain selectively binds to an HIV antigen.
