The apparatus for nucleic acid extraction includes: a first rack having a plurality of sample tube receivers arranged in a circle; a second rack having a plurality of elution tube receivers arranged in a circle on an outer side thereof and a washing solution receiver positioned at the center thereof, a part of the washing solution receiver extending outwards to have projections formed in alternation with a plurality of the elution tube receivers; a main body having the first and second racks arranged to position the first rack on the top of the second rack; a rotational driver for separately rotating the first and second racks; a dispenser for separately dispensing a washing solution and an eluting solution into sample tubes; and a pressurizer for maintaining the inside of the sample tubes under raised pressure.

Multi-stage purification method and apparatus of extracellular vesicles using aqueous solution two-phase system

This disclosure relates to a multi-stage method and apparatus for purifying extracellular vesicles using an aqueous two-phase system, in which extracellular vesicles mixed and contaminated with proteins can be isolated and purified in a large amount at high purity within a short time through a multi-stage purification process using an aqueous two-phase system, thereby removing 95% or more of proteins and obtaining high-purity extracellular vesicles, resulting in very high processing efficiency compared to conventional techniques. In particular, the method of the disclosure does not require an expensive device or material such as an ultracentrifuge or an antibody, and can be performed at low cost and is thus economical and highly competitive. Furthermore, the extracellular vesicles thus isolated and purified can be employed in analysis methods such as RT-PCR or western blot, and can be utilized for research fields and disease diagnosis using the same.

Disclosed are methods and devices for lysing cells to release extract genomic DNA (gDNA). The methods use a mixture of microscopic glass beads and cells (for example, spores) that form a semi-dry cake that clings to a larger metal ball and the sides of the tube during bead beating lysis, greatly improving the efficiency of the bead beating process. The devices produce a chaotic motion which ensures that sufficient force is generated to open the cells, and that the metal ball impacts are distributed across the interior surface of the container so that all of the cell mixture is subjected to sufficient impacts to break the cells. As a result, spores and other difficult-to-lyse microbes, can be opened in seconds. The method reduces the number of steps and hands-on time by rapidly opening difficult to lyse cells, while preserving the integrity of the DNA.

The system is closable to the atmosphere, and includes a cell suspension collection tube containing gel and anticoagulant, adapted such that, when containing cell suspension and centrifuged after treatment the collection tube yields separation fractions, a first fraction comprising cellular fluid, a second fraction comprising gel layer, a third fraction comprising enriched cellular portion on top of the gel layer and a fourth fraction comprising a cell poor portion. The cell poor portion may be extracted and discarded prior to extracting the enriched cellular portion.
