The method includes the follows: using a dedicated amplification medium of immune cells to perform first-stage amplification culture on mononuclear cells to obtain preliminarily amplified immune cells; using a dedicated induction medium of immune cells to perform second-stage induction and amplification culture on the preliminarily amplified immune cells to obtain induced immune cells; using a dedicated activation medium of immune cells to perform third-stage activation and amplification culture on the induced immune cells to obtain a large number of immune cells with activation functions; using a dedicated cryopreserving fluid of immune cells to cryopreserve the immune cells to obtain cryopreserved immune cells; and performing preservation according to ABO/RH typing and HLA typing; and establishing an information file of immune cells for retrieval to construct an immune cell bank.

The present invention relates to a method, which patterns 3D organoids prepared from human pluripotent stem cells and chops the same so as to culture oligodendrocyte progenitor cells, and induces the differentiation thereof so as to obtain a large quantity of finally differentiated oligodendrocytes. Compared to cells differentiated by a conventional differentiation method, oligodendrocytes obtained in a large quantity have the same or superior reproducibility, stability, and functionality and have remarkably shortened differentiation time, and thus are expected to be very useful for cell therapeutic agents or for screening for therapeutic drugs.

Donor cells may include, for example, a precursor cell such as an embryonic stem cell or other precursor cell. The disclosed methods use synchronization conditions to produce a synchronized pooled-precursor cell population, which may then be differentiated into an organoid composition. The primary gynecological tumor cell culture obtained by the method of the present invention can be used for various cell-based in vitro experiments, second-generation sequencing, construction of animal models, construction of cell lines and the like. It is foreseeable that this culture method has broad application prospects in the fields of gynecological tumor research and clinical diagnosis and treatment.

Hormone responsive tissue culture system and uses thereof

The invention provides tissue culture system for primary cells (e.g. normal mammalian primary epithelial progenitors). This system includes: a) a serum-free, chemically defined cell culture media; and, b) methods for isolation and in vitro long-term propagation of primary cells (e.g. primary epithelial cells). Primary cells so isolated and cultured can be kept undifferentiated and proliferate for many weeks (>15 weeks) or population doubling (>35 PD) without senescence, or any detectable genetic alterations. Upon changing media/culture conditions, these cells can be induced to differentiate. The invention also provides methods to transform normal primary cells so cultured into “cancer stem cells.” The genetically defined cancer stem cell tumor model mimics the behavior of the disease closely, e.g., the cells are invasive, hormone responsive and metastatic when injected into mice. The tumor cells express genes that are specific to cancer stem cells identified in patient samples.
