Novel RNA Composition and Production Method for Use in iPS Cell Generation
This invention generally relates to a novel RNA composition and its production method useful for generating and expanding induced pluripotent stem cells (iPS cells; iPSC) as well as adult stem cells (ASC). The RNA composition so defined can be used for producing not only non-transgenic but also tumor-free iPS cells. The defined RNA composition contans at least two types of different RNA constructs; one is “miR-302 precursor RNA (pre-miR-302)” and the other is “RNA-dependent RNA polymerase (RdRp)” mRNA. Both of pre-miR-302 and RdRp mRNA contain highly structured RNA comformations, such as hairpin and stem-loop structures. To produce highly structured RNAs, a novel PCR-IVT methodology has been developed and used with a specially designed RNA polymerase-helicase mixture activity.

Method for inducing differentiated cell into Mesenchymal Stem Cell, and combinations of regulatory targets thereof
A method for inducing differentiated cells into mesenchymal stem cells (MSCs), and combinations of regulatory targets thereof. The method includes performing a directional induction on the differentiated cells to prepare the mesenchymal stem cells. The directional induction includes treating cells by inhibiting the TGF-β signal pathway, inhibiting the activity of PKC, activating the WNT/β-catenin signal pathway and activating the cAMP signal pathway. By regulating corresponding signal pathways and/or enzymatic activities by stages, the differentiated cells are induced into the mesenchymal stem cells.
Method for activating Oct4 for induction of pluripotent stem cells
The invention relates to a method for inducing pluripotent stem cells, wherein a mammalian cell is contacted with a compound characterized by a general formula 1 wherein R.sup.1 is (CH.sub.2).sub.mE, with E being CCH or CN and m being 0, 1, or 2, and R.sup.2 is selected from F, CI, Br, OR.sup.3 and R.sup.3, with R.sup.3 being selected from H, (CH.sub.yF.sub.2-y).sub.n CH.sub.XF.sub.3-X, with n=0 or 1. The invention further relates to stem cells obtained by the method of the invention, and culture media comprising the compound of the invention.
