Marker genes for oocyte competence
Cumulus cell (CC) gene expression is being explored as an additional method to morphological scoring to choose the embryo with the highest chance to pregnancy. The present invention relates to a novel method of identifying biomarker genes for evaluating the competence of a mammalian oocyte in giving rise to a viable pregnancy after fertilization, based on the use of live birth and embryonic development as endpoint criteria for the oocytes to be used in an exon level analysis of potential biomarker genes. The invention further provides CC-expressed biomarker genes thus identified, as well as prognostic models based on the biomarker genes identified using the methods of the present invention.
METHODS OF USING ZSCAN4 FOR REJUVENATING HUMAN CELLS
The present disclosure relates to methods for increasing telomere length in one or more human cells and/or increasing genome stability of one or more human cells, for example by contacting one or more human cells with an agent that increases expression of Zscan4 in the one or more human cells. Methods of treating a subject in need of telomere lengthening, treating a disease or condition associated with a genomic and/or chromosome abnormality, of rejuvenating one or more human cells, of rejuvenating tissues or organs, and of rejuvenating a subject in need thereof, for example by contacting one or more human cells in the subject with an agent that increases expression of Zscan4, or by administering to a subject in need thereof, an agent that increases expression of Zscan4 are also provided.
Agent and method for enhancing fertility
An agent capable of promoting proliferation and differentiation of granulosa cells is disclosed which comprises a growth and differentiation factor-9 (GDF9) protein comprising a modified GDF9 polypeptide monomer which includes at least one amino acid substitution that enhances binding to and/or activation of activin-like kinase 4 and/or 5 receptor (ALK4/5). The agent is preferably provided in a mature dimeric form (eg comprising two monomers of the same modified GDF9 polypeptide monomer) and/or in a pro/mature complex form. The agent may be suitable for, inter alia, promoting oocyte maturation in vitro for use in assisted reproductive technologies.
Nuclear transfer
The present invention provides methods and compostions to improve the efficiency of somatic cell nuclear transfer (SCNT). There is increasing evidence that the epigenetic state of donor nuclei has a significant impact on potential of nuclear transfer embryos to develop into blastocysts, from which pluripotent stem cells are derived. Strategic application of histone agents, capable of altering epigenetic state such as methylation, allows zygotic activation and robust blastocyst generation.
Compositions and methods for maturation of oocytes in vitro
The present invention relates to culture media for oocytes and uses thereof. Specifically, media for culturing an oocyte in vitro are disclosed, wherein said media comprise granulocyte macrophage-colony stimulating factor (GM-CSF). The presence of GM-CSF in the media increases the maturation and/or developmental competence of the oocyte making it suitable for use in subsequent assisted reproductive technologies. Methods for increasing the maturation and/or developmental competence of an oocyte are also disclosed.
Systems and methods for natural cryoprotectants for preservation of cells
Embodiments of the present invention provide plant-derived extracts as a replacement for traditional cryoprotectants used to freeze tissue and cells providing a method to decrease post-thaw damage as created by the cryoprotectant. For example, extracts from the genus Hippophae or other plant sources or compositions may be used as a cryoprotectant or may even be used to replace at least some of a traditional cryoprotectant.

The present inventions provide methods to control the heterogeneity of Fc-containing proteins, such as antibodies produced in cell culture, particularly mammalian cell culture by controlling culture pCO.sub.2, as well as products produced by these methods. Among other things, the inventions provide for lowering the percentage of acidic charge variants in antibody products. Proteins that comprise Fc moieties include but are not limited to Fc-containing proteins, such as antibodies and antibody derivatives, and fragments of both.

A method for inducing immature oocytes includes introducing four genes consisting of FIGLA, NOBOX, LHX8 and TBPL2, or transcripts or expressed proteins thereof, into at least one type of cell selected from the group consisting of pluripotent stem cells, epiblast-like cells and primordial germ cells. A method for producing mature oocytes includes introducing four genes consisting of FIGLA, NOBOX, LHX8 and TBPL2, or transcripts or expressed proteins thereof, into at least one type of cell selected from the group consisting of pluripotent stem cells, epiblast-like cells and primordial germ cells, and co-culturing the cell obtained after the introduction and ovarian somatic cells.
