A computational microscope and a method for its operation are disclosed. In some embodiments, the microscope maps points on a sample to point in an intensity pattern on a one-to-many basis. The microscope utilizes illumination angle coding, polarization coding, amplitude coding, and phase coding to capture more information than prior art computational microscopes. Although the resulting intensity patterns are not human-interpretable images of the sample, they contain more information about the sample, by virtue of the aforementioned coding techniques, than is captured by prior-art microscopes. Machine-learning algorithms, such as neural networks, are used to analyze the intensity patterns and extract useful information, such as cellular events or cell behavior.

In order to be able to observe also non-fluorescent regions in the tissue (3) without complicated filter arrangement, the medical device (1) according to the invention comprises at least one filter system (16, 38) which comprises, in a filter plane (18), comprises a filter area (20) and a transmission window (22). The filter area (20) comprises a band pass filter (24) having at least one passband (44) comprising the at least one excitation waveband. The transmission window has a passband (48) which is wider than the passband (44) of the filter area (20). In particular, a filter layer (64) of the filter area (20) may be missing in the transmission window (20).

The plurality of transparent windows is configured to increase the transmittance of light through the display to the sensor. The transparent windows may have non-periodic portions to mitigate diffraction artifacts in light that passes through the display to the optical sensor. The transparent windows may be shifted by a random amount in a random direction relative to a grid defining point and/or may be randomly rotated to increase the non-periodicity. A transparency gradient may be formed between the transparent windows and the surrounding opaque portion of the display. The transparent windows may be defined by non-linear edges.
