An application of a branched-chain α-ketoacid dehydrogenase complex in preparation of malonyl coenzyme A. A method for preparing malonyl-CoA using a branched-chain α-ketoacid dehydrogenase complex, the method comprising introducing a gene encoding a branched-chain α-ketoacid dehydrogenase complex into a biological cell strain to obtain a recombinant cell strain capable of expressing the gene encoding the branched-chain α-ketoacid dehydrogenase complex; culturing the recombinant cell strain to prepare malonyl-CoA; the branched-chain α-ketoacid dehydrogenase complex is the following M1) or M2): M1) a set of proteins consisting of a bkdF protein, a bkdG protein, a bkdH protein and a lpdA1 protein; M2) a set of proteins consisting of a bkdA protein, a bkdB protein, a bkdC protein and the lpdA1 protein. Experimental results show that by using the branched-chain α-ketoacid dehydrogenase complex, not only malonyl-CoA can be prepared, but also a target product using malonyl-CoA as an intermediate product can further be prepared.

Method for producing tetrahydropyrimidine by fermenting recombinant Corynebacterium glutamicum
11512333 · 2022-11-29 · ·

A method for producing ectoine by fermenting recombinant Corynebacterium glutamicum. The recombinant Corynebacterium glutamicum is obtained by overexpressing, in Corynebacterium glutamicum, an aspartokinase gene lysC of which feedback inhibition is relieved, then replacing the promoter of the dihydrodipicolinate synthase in the recombinant bacterium to attenuate the activity of the dihydropyrimidine dicarboxylic acid synthase, and then transforming the recombinant bacterium with the ectoine synthetic path related gene ectABC. The recombinant Corynebacterium glutamicum can be fermented using different cheap raw materials under a low salt condition to produce ectoine, and use cheap corn slurry instead of expensive yeast powder as a nutritional component, so as to further reduce the costs of the raw materials. In addition, the recombinant Corynebacterium glutamicum solves the biosafety problem, simplifies the post-extraction process, and has a good market application prospect.

Bacillus thuringiensis RTI545 compositions and methods of use for benefiting plant growth and controlling plant pests

Compositions are provided that include a new Bacillus thuringiensis strain designated RTI545 for use in benefiting plant growth and controlling plant pests. In particular, the RTI545 strain is useful for controlling plant nematode, insect and fungal pests. The compositions include plant seeds coated with the RTI545 strain. The compositions can be applied alone or in combination with other microbial, biological, or chemical insecticides, fungicides, nematicides, bacteriocides, herbicides, plant extracts, plant growth regulators, or fertilizers. In one example, enhanced growth and insect control are provided by delivering at the time of planting a combination of a chemical insecticide such as bifenthrin and a liquid fertilizer to plants or seeds treated with RTI545.

Endophytic microbial strains as biocatalysts isolated from fresh plant samples, their compositions, and methods of use thereof to enhance the growth and/or yield of a plant in the presence of reduced (synthetic or otherwise) nitrogen and phosphorus fertilizers are provided. The selected endophytic microbial strains serve as biocatalysts to solubilize organic (proteinaceous) nitrogen otherwise unavailable to plants for their nutritional needs. In addition, selected endophytic microbial strains serve as biocatalysts to solubilize mineral-P and mineralize organic-P otherwise unavailable to plants for their nutritional phosphate needs. Thus defined, biocatalysts will serve to replace or reduce the requirement of (synthetic or otherwise) nitrogen and phosphorus fertilizers. Also provided are materials and methods for inoculating plants with these biocatalysts at carefully selected inoculum densities to reliably reduce the amount of nitrogen and phosphorus fertilizers by 30-50% thus accomplishing optimal yields in a cost-effective manner.
