METHOD AMD STRAINS FOR REDUCING BYPRODUCT FUMARIC ACID IN FERMENTATION PROCESS OF L-MALIC ACID AND USE THEREOF
20230167471 · 2023-06-01 ·

The disclosure discloses an Aspergillus niger engineered strain for reducing byproduct fumaric acid in a fermentation process of L-malic acid. The Aspergillus niger engineered strain is an Aspergillus niger engineered strain in which a fumarate hydratase gene fum is knocked out. The disclosure overcomes the defects in the prior art, in the current process of producing malic acid through fermentation of Aspergillus niger, byproduct fumaric acid can be accumulated with the generation of malic acid so as to cause the improved cost of the subsequent malic acid purification process. The disclosure provides an Aspergillus niger engineered strain in which a fum gene is knocked out and a method for greatly reducing byproduct fumaric acid in the fermentation production of Aspergillus niger.

TRANSFORMED FUNGUS HAVING ENHANCED ERGOTHIONEINE PRODUCTIVITY AND METHOD FOR PRODUCING ERGOTHIONEINE

The purpose of the present invention is to provide an organism having an ergothioneine productivity that is capable of easily producing ergothioneine within a short period of time at a high yield, as compared with a conventional technology, and, therefore, enables ergothioneine production on an industrial scale. This purpose can be achieved by a transformed fungus into which a gene encoding enzyme (1) or genes encoding enzymes (1) and (2) have been inserted and in which the inserted gene(s) are overexpressed. (1) an enzyme catalyzing a reaction of synthesizing hercynyl cysteine sulfoxide from histidine and cysteine in the presence of S-adenosyl methionine, iron (II) and oxygen. (2) An enzyme catalyzing a reaction of synthesizing ergothioneine from hercynyl cysteine sulfoxide using pyridoxal 5′-phosphate as a coenzyme.

Synthesis of furans from sugars via keto intermediates

The present invention provides a method of preparing a furan derivative comprising the steps of (a) converting a monosaccharide to provide a keto-intermediate product; and (b) dehydrating the keto-intermediate product to provide a furan derivative; wherein the keto-intermediate product is pre-disposed to forming keto-furanose tautomers in solution. The method may further comprising a step of oxidizing the furan derivative to provide a furandicarboxylic acid or a furandicarboxylic acid derivative.

The present disclosure discloses a visualized screening method for an Aspergillus recombinant strain with multigene editing and belongs to the technical field of gene engineering. CRISPR-Cas9 is used in the disclosure to cleave spore color change-related genes and a target gene in Aspergillus at the same time, such that editing of the target gene is visualized and an Aspergillus niger strain with multigene editing can be rapidly and efficiently screened out through spore phenotypes. Through different combinations of visualized genes and non-phenotypic change genes, rapid screening of the strain with multigene editing and simultaneous screening of multiple visualized genes are realized, and use of resistance genes in industrial strains is reduced.

Method for the desalination of seawater using solar energy

A technique to desalinate seawater using melanin-concentrated solar energy wherein the melanin is extracted from a local isolate Aspergillus niger. A device consists of two fixed upper and lower containers with same volume of seawater in both, with or without melanin powder dissolved in the lower container at rate of 0.17 gm of melanin powder per 10 ml of water. The device is put outdoors under direct sunlight during daytime, circular water droplets free of salt starts to appear on the external bottom of upper container. Water droplets are collected by a sterile glass rod, pH of droplets water is about 7.1. Yield of fresh water is approximately 10 ml droplets water from 600 ml seawater per hour; after 24 hours day and night incubation, seawater in the upper container dries out leaving salt crystals. Yield of 1000 m3 seawater is 100 m3 freshwater (1000 L seawater yield 100 L freshwater).
